Yes, I think he was a real person before he entered the Black Lodge. The other "spirit" Mike, implied that in one of the early episodes.
I think Bob actually had a tendency to hate women while he was human (particularly a certain type, e.g. women like Laura Palmer and Madeline Ferguson), but the Black Lodge took that hatred to a new level by releasing his evil to it's fullest extent and making him into the killer he became.
