I would say Groucho Marx. Because Bugs Bunny was based on Groucho Marx (his jokes, character and mannerisms) and nearly every modern comedian will have been influenced by Bugs Bunny as a child. There are also the direct influences of Marx himself and the films he appeared in.
Because Groucho always played the same character in films and shows it meant that the character was developed over a period of decades and as such wuld have a greater influence than one who has only been around for a few years.
It is the influence of the Groucho character that can be seen in the style of so many modern stand up and sitcom characters that make him so influential.

My second choice would have to be Homer Simpson, and my third being Father Dougal.

I've got to go with either Peter Griffin of Family Guy, Stewie Griffin of Family Guy, or Dave Chappelle the stand-up comic.

Dave Chappelle's "Rick James" episode will probably go down in history as the funniest skit ever done.

But Peter and Stewie Griffin are constantly funny; they never reached the pure hilariousness of the Rick James skit, but they are a steady source of comedy every episode of Family Guy.

I tried hard to think of someone else because whenever the do this sort of survey on TV,usually ch4 or UKGold,Only Fools and Horses always wins and it gets a bit boring but the thing is when I think about it I realise it probably was the best comedy programme ever.

Oh man it's got to be the witch skit from Monty Python's Quest for the Holy Grail. "She turned me into a newt. I got better." OR Monty Python's And Now for Something Completely Different with the musical mice.
