Well, their relationship has always been bumpy. They were put under a spell to love each other in "Something Blue" (season 4). But I think they became friends in "Intervention" (season 5) when Spike withstands Glory's torture and refuses to tell her who the key is. After that, Buffy respects him more. And then they kiss for the first time in "Once More, With Feeling" (season 6) and have sex for the first time a little later. But it's the seventh season where they really fall in love with each other.
