There is no way to be 100% certain why it is happened but many people do believe it is because John Lennon mooned Paul (pulled down his pants and flashed his bare ****).

The story is that while Paul sings the lyric "so he waits behind" John mooned him from inside the recording booth. That would be a fair explanation of why Paul laughed.

Some people claim that John Lennon was not at the sessions for the recording of this song because he was recovering from a car accident.

So we will never know exactly why, but I like the mooning story. It could very well be true because it is not out of character for Lennon as we all know.
I have no idea...I never really noticed it. That song used to creep me out when I was a kid ?
I might be remembering wrong, but I think John mooned him while they were in the studio, so he laughed. I guess they thought it was a nice touch and kept it in. I might be thinking of a different song.
It's a funny song (in a dark and offbeat way) and they probably had to do a lot of takes. I imagine there were some where they had to stop and start again because they were cracking each other up. Once a case of giggles gets started it's hard to stop.
McCartney can be heard sniggering as he sings the line "... writing fifty times I must not be so ..." on the studio recording. This is often rumored to be because as McCartney sang the line "so he waits behind," Lennon mooned McCartney from the control room. This is untrue as Lennon was not present at the recording sessions for the song.
