First the kryptonite will slow him down and hamper his powers slowly draining him of not only his powers given to him by the sun but also his lifeforce eventually it will kill him with prolonged exposure, though theres the variable of how potent the piece is some are not as powerful as others though they all posses the same effect.
it would proably have to pierce his skin and be get into his system and be there for a couple of minutes before it killed him
