This is a hard thing to explain with simple words but I will try.

Guitar scale is about the distance between the string nut and the bridge saddles. It is best to measure between nut and center of 12th fret and multiply by 2.

For example, most Gibson Les Paul have a 24 5/8" (625mm) scale, this means that the distance from the nut to 12th fret is 12 5/16" (312,5mm), but the distance between nut and bridge saddles is 24 3/4" (629mm). This is because a small amount is added to scale length called "compensation" to allow the guitar to play in tune with itself.

Other guitars have different scale lengths. If the scale gets longer then you get a thinner sound and more sustain. If you get a shorter scale the sound becomes deeper and less sustain.

When building a guitar the combination of scale length, materials, and construction go together to make the sound of the guitar.
