My favorite Joker is Mark Hamill's Joker. He does the voice of the Joker for Batman the Animated Series and I always thought he was the best!
I'm a big fan of Cesar Romero as the joker in the 1960s Batman TV series. It's a bit of a light-hearted and goofy portrayal, to be sure, but that is in keeping with the series. I love his maniacal laughter.
Another good one is the Laughing Man in the Batman: Nosferatu comic, my favourite of the few comics I've read. I think Heath Ledger looks promising. It should be a good movie.

I have to say that Jack Nicholson did the best job creating the appropriate image of the Joker.
